What it is studied for

A single peptide acting at both the GIP and GLP-1 receptors, an approach that produced larger effects in metabolic models than GLP-1 agonism alone. The standard dual-agonist comparator in incretin research.

About Tirzepatide

Tirzepatide is a synthetic 39-amino-acid peptide built on the native GIP sequence rather than on GLP-1, which is what separates it from semaglutide and the other selective GLP-1 agonists in this catalogue. Thirty-seven of its residues are standard amino acids; positions 2 and 13 carry aminoisobutyric acid, a non-coded residue that resists the enzymatic cleavage which limits how long native incretins survive.

A C20 fatty diacid is attached partway along the chain. That modification binds the peptide to serum albumin, and it is the reason the molecule persists long enough for weekly rather than daily administration in the study protocols where it appears.

It was first characterised as LY3298176 by Coskun and colleagues at Eli Lilly in 2018. Later receptor-level work described its activity as imbalanced and biased — markedly more potent at the GIP receptor than at GLP-1, and recruiting β-arrestin differently from either native ligand. That asymmetry, rather than simple dual agonism, is the property most incretin research now examines it for.

Handling

Bring the vial to room temperature before opening, add diluent slowly down the inside wall rather than onto the powder, and let it dissolve undisturbed — no shaking. Store refrigerated on arrival, and refrigerate the solution once reconstituted.

Questions

How is Tirzepatide supplied?

Dual GIP and GLP-1 receptor agonist, 10 mg per vial. It ships in unbranded outer packaging that lists no contents. Store refrigerated on arrival. The lyophilised powder is stable in transit at ambient temperature.

How is Tirzepatide reconstituted?

Bacteriostatic water is the usual diluent. Run it down the inside wall of the vial rather than onto the powder and let it dissolve without shaking. With 10 mg in the vial, the diluent volume you choose sets the final concentration. The reconstitution calculator on this site works out the draw volume for any combination.
