About CagriSema

This vial holds two different peptides, 5 mg of each, and they come from unrelated hormone families. Semaglutide is a GLP-1 analogue, acting on the incretin axis. Cagrilintide is an amylin analogue, acting on a pathway that has nothing to do with GLP-1 — amylin is co-secreted with insulin from the same beta cells, but signals through its own receptors.

What makes the pairing coherent is not their mechanism but their chemistry. Both carry a fatty diacid chain attached through a gamma-glutamate spacer, which binds them reversibly to serum albumin. Two molecules from separate families, modified the same way, ending up with matched persistence — which is what lets them be given on the same schedule from one vial at all.

The research question behind co-formulation is whether two satiety pathways are additive or whether they overlap and one masks the other. A blend cannot answer that on its own: with both compounds fixed in a 1:1 ratio by a shared reconstitution volume, neither can be varied independently.
